Mix and Heat:
Combine the agar agar powder and other dry ingredients in a heat-resistant container.
Add distilled water and stir to dissolve the ingredients.
Heat the mixture on a stove or in a microwave until it boils, stirring occasionally to ensure all ingredients are fully dissolved.
Sterilize:
Pour the hot agar mixture into media bottles and autoclave at 15 psi (103.4 kPa) for 20 minutes to ensure all contaminants are killed.
Pour Plates:
Allow the sterilized agar mixture to cool to 50⁰C.
In a sterile environment, pour the agar into sterile petri dishes, filling each dish with 20-25ml.
Cover the petri dishes immediately to prevent contamination.
Solidify and Store:
Let the agar plates cool and solidify at room temperature.
Once solidified, store the plates in a refrigerator or cool, dark place until ready for use. Use within a reasonable time frame to ensure sterility.
